Clay minerals are suitable matrices to anchor organic molecules such as antimicrobial peptides (AMPs) so that their bioactivity is maintained, enabling the formation of new materials with potential for new applications in biotechnology. The objective of the present study was to develop a nanostructured flm where the properties of palygorskite (Plg) were combined at the molecular level with Dermaseptin 01 (DRS 01), in which the clay mineral also served as a substrate for the immobilization of this peptide. The flms were prepared using the Layer-byLayer (LbL) self-assembly technique. Crude palygorskite without purifcation (Plg IN) was subjected to physical and chemical procedures to increase its adsorptive properties. The structure, chemical composition, and morphology of Plg were investigated by X-ray difraction (XRD), Fourier-transform infrared spectroscopy (FTIR), X-ray fuorescence spectrometry (XRF), scanning electron microscopy (SEM), and transmission electron microscopy (TEM). LbL flms were adsorbed onto ITO (Indium Tin Oxide) and characterized electrochemically by cyclic voltammetry (CV), UV-Visible spectroscopy, and atomic force microscopy (AFM). For the ITO/DRS 01 and ITO/Plg/DRS 01 flms, an oxidation process at +0.77 V was observed, confrming that the DRS 01 maintained its electroactive behavior and intrinsic properties. The results also showed that Plg served as excellent support for the immobilization of DRS 01, increasing its concentration and availability in the flm form. This work reported immobilizing the DRS 01 peptide with Plg for the frst time in an ultrathin flm with bioactive properties. Thus, the flm developed can be explored for applications such as biosensor devices and antimicrobial coating materials as well as other biotechnological applications. O objetivo deste estudo foi desenvolver e avaliar as características microbiológicas e sensoriais aceitação doce de leite de búfala com adição de morango. O leite foi filtrado e adicionado 1,8 g de bicarbonato de sódio, aquecido a 90 ° C e homogeneizado com 600 g de açúcar refinado, por 120 minutos, para ficar cremoso. Depois ele juntou o doce 50g de morango tratado e picado. O derivado foi embalado a quente, resfriado e armazenado a 10°C por análises microbiológicas e sensoriais subsequentes. A análise microbiológica resultou em 2x10 MPN/g para coliformes a 45 º C, <1x10 UFC/g para Staphylococcusaureus e ausência de Salmonella em 25g de leite fresco, o que indica cumprimento das normas vigentes legislação e eficiência nos procedimentos higiênicos adotados no manuseio. A derivada foi bem aceito pelos jurados, com nota média de 8,4. Observe que a alta aceitabilidade do derivado pode estar relacionada ao sabor de morango, que provavelmente contribuiu para sua palatabilidade. O doce preparado com leite de búfala sabor morango pode ser produzida por pequenos agricultores porque utiliza tecnologia de fácil acesso, tornando-a opção de alto valor, incluindo alimentação de populações e merenda escolar |
